Multi-column chromatography (MCC) has gained popularity for monoclonal antibody (mAb) capture processes, potentially offering benefits of higher productivity and lower resource consumption, including buffers and resin. However, there is still a need to demonstrate the efffectiveness of polishing steps using MCC. This study will do just that by demonstrating the implementation of step gradients for mAb polishing using a MMC system and technique.

Recent shifts in mAb production favor continuous over batch processes, yet downstream processing remains a costly bottleneck. Multi-column chromatography (MCC), including continuous capture and simulated moving bed (SMB) chromatography, offer cost-effective alternatives with higher productivity and lower resource consumption. This study demonstrates the implementation of step gradients for mAb polishing, further extending the use-case spectrum of SMB technology in biopharmaceutical manufacturing.
